Molecular cloning and characterization of a novel gene encoding tetraspanin of Schistosoma japonicum

چکیده
In this study, a novel tetraspanin-like gene product (Sj04/ TM4) was identified. An EST named JAYN004.GYL (Gen Bank accession number AW231251) matched the Sj25 gene of Schistosoma japonicum with 97 % (539/554) identity. The longest cDNA insert (1079 bp) was obtained from the recombinant clone JAYN004.GY by PCR. Sequence analysis showed that it contains a complete open reading frame (ORF) coding for a protein of 224 amino acids with a predicted molecular mass of 25,622.58 Dalton. Alignment analysis with the entire ORF revealed that it had 93.3 % identity in 224 aa overlap % identity to Sj25/TM from S. japonicum. It also exhibited 31.4 % in 220 aa overlap and 21.1 % in 209 aa overlap, respectively to tetraspanin TE736 from S. japonicum and Sm23 antigen from S. mansoni, whose are belong to the members of tetraspanins. Analysis of amino acid sequence showed that this gene product contains all the conserved subdomains found in other members of tetraspanin superfamily. Hydropathy analysis of the deduced amino acid sequence revealed a profile very similar to those of other tetraspanins containing 4 transmembrane domains. Taken together, Sj04/TM4 is a novel gene encoding tetraspanin-like protein, or belongs to a subtype of Sj25/TM4 tetraspanin superfamily.

Cloning and molecular characterization of TaERF6, a gene encoding a bread wheat ethylene response factor
Ethylene response factor proteins are important for regulating gene expression under different stresses. Different isoforms for ERF have previously isolated from bread wheat (Triticum aestivum L.) and related genera and called from TaERF1 to TaERF5. We isolated, cloned and molecular characterized a novel one based on TdERF1, an isoform in durum wheat (Tri...
متن کاملMolecular cloning and tissue distribution of a Schistosoma japonicum gene encoding AMY-1.
It is well known that the mammalian associate of Myc-1 (AMY-1) plays a significant role in spermatogenesis or cellular differentiation. A full-length complementary DNA (cDNA) encoding AMY-1 of Schistosoma japonicum (SjAMY-1) was identified and isolated from a cDNA library. The gene contained an open reading frame of 315 nucleotides, encoding 105 amino acids. Sequence analysis showed that SjAMY-...
متن کاملInconsistent Protective Efficacy and Marked Polymorphism Limits the Value of Schistosoma japonicum Tetraspanin-2 as a Vaccine Target
BACKGROUND Schistosoma mansoni tetraspanin 2 (Sm-TSP-2) has been shown to be strongly recognized by IgG1 and IgG3 antibodies from individuals putatively resistant to schistosome infection, but not chronically infected people, and to induce high levels of protection against challenge infection in the murine model of schistosomiasis. Amplification by PCR of homologous sequences from male and fema...
متن کاملCloning and Characterization of cbhII Gene fromTrichoderma parceramosum and Its Expressionin Pichia pastoris
The genomic and cDNA clones encoding cellobiohydrolase II (CBHII) have been isolated and sequenced from a native Iranian isolate of Trichoderma parceramosum, a high cellulolytic enzymes producer isolate. This represents the first report of cbhII gene from this organism. Comparison of genomic and cDNA sequences indicates this gene contains three short introns and also an open reading frame codin...
متن کاملMolecular characterization of S. japonicum exosome-like vesicles reveals their regulatory roles in parasite-host interactions
Secreted extracellular vesicles play an important role in pathogen-host interactions. Increased knowledge of schistosome extracellular vesicles could provide insights into schistosome-host interactions and enable the development of novel intervention strategies to inhibit parasitic processes and lessen disease transmission. Here, we describe biochemical characterization of Schistosoma japonicum...
